However, the exact pathogenesis of Alzheimer’s remains elusive, and existing highly recognized pathogenesis includes the amyloid cascade hypothesis, Tau neurofibrillary tangles hypothesis, and neuroinflammation hypothesis. The major diagnoses of Alzheimer’s disease include neuroimaging positron emission computed tomography, magnetic resonance imaging, and cerebrospinal fluid molecular diagnosis. The therapy of Alzheimer’s disease primarily relies on drugs, and the approved drugs on the market include acetylcholinesterase drugs, glutamate receptor antagonists, and amyloid-β monoclonal antibodies. Still, the existing drugs can only alleviate the symptoms of the disease and cannot completely reverse it. This review aims to summarize existing research results on Alzheimer’s disease pathogenesis, diagnosis, and drug therapy, with the objective of facilitating future research in this area.

Such changes in premotor inputs can contribute to homeostatic imbalance of motor neurons. Here, we show that the Extended Synaptotagmin 1 (Esyt1) presynaptic organizer is downregulated in V1 interneurons. V1 restricted overexpression of Esyt1 rescues inhibitory synapses, increases motor neuron survival, and ameliorates motor phenotypes. Two gene therapy approaches overexpressing ESYT1 were investigated; one for local intraspinal delivery, and the other for systemic administration using an AAV-PHP.eB vector delivered intravenously. Improvement of motor functions is observed in both approaches, however systemic administration appears to significantly reduce onset of motor impairment in the SOD1G93A mice in absence of side effects. Altogether, we show that stabilization of V1 synapses by ESYT1 overexpression has the potential to improve motor functions in ALS, demonstrating that interneurons can be a target to attenuate ALS symptoms. The overexpression of Esyt1 presynaptic organiser in V1 spinal interneurons restores connectivity, increases motor neuron survival, and ameliorates motor phenotypes in ALS mice. So, inhibitory interneurons are a target to attenuate ALS symptoms.

Cognitive inhibition enhances the selection of task-relevant stimuli and is closely related to neural changes that occur across the lifespan. Since few studies have focused on the entire lifespan, this study aimed to assess cognitive inhibition abilities in a sample of 425 healthy participants (age range: 7–88 years) using the Stroop task. The participants were grouped according to age into children, adolescents, young adults, adults, middle-aged adults, and older adults. A series of ANOVAs considered Group as the independent variable and Performance indices as the dependent variables. The children did not show an interference effect (Stroop effect), likely due to the lack of an automated reading process as a consequence of ongoing brain maturation. Adolescents and young adults performed significantly faster than older adults did. The results indicate that response speed reaches its peak during adolescence and young adulthood and then slightly decreases until older age. Nevertheless, when compared with the other groups, only older adults showed significant differences in the Stroop effect, suggesting that inhibitory abilities remain relatively consistent throughout adulthood but rapidly worsen in recent years due to the physiological decline in cognitive and brain functioning associated with aging.

Freesurfer 6 was applied to generate the morphometry of brain regions. A multivariate pattern analysis was used to derive brain morphometry patterns associated with food addiction. General linear modeling and mediation analyses were conducted to examine the effects of ELA and resilience on food addiction in individuals with obesity. Statistical significance was determined at a level of p < 0.05. High levels of ELA showed a strong association between reward control brain signatures and food addiction (p = 0.03). Resilience positively mediated the effect of ELA on food addiction (B = 0.02, p = 0.038). The study was conducted in an Arabica coffee plantation. The study consisted of eight treatments and four replications in a 2 × 2 × 2 factorial scheme: two flight heights (1.5 and 3.0 m), two spray tips (hollow cone and flat fan), and two spray volumes (10 and 15 L ha−1). Deposition was assessed by detecting a tracer in the coffee leaves and fruit using spectrophotometry. The coffee berry borer-control efficacy trial was conducted in two areas by evaluating the percentage of damaged fruit 60 days after two insecticide applications. The flight height of 1.5 m promoted higher spray deposition on leaves and fruit and a lower incidence of damaged fruit. Flat fan spray tips resulted in higher spray deposition on the leaves, not interfering with the deposition on fruit or the coffee berry borer control. Increasing the spray volume from 10 to 15 L ha−1 did not increase spray deposition on coffee leaves and fruit. Chlorpyrifos applied via UAVs reduced the incidence of coffee berry borer.
